In this lecture we will give an overview of recent developments in targeted and untargeted metabolomics and lipidomics analysis. Pitfalls and challenges for implementing metabolomics and lipidomics in drug discovery will be discussed. Showcasing specific examples, the use of lipidomics and metabolomics for target discovery, mode of action and physiological response marker elucidation will be explained. Next to a general introduction to the topic we will specifically focus on non-alcoholic steatohepatitis (NASH) and the use of lipidomics technologies. Showing data from one of our chemical probes in a humanized preclinical mouse model we will discuss the application of lipidomics and metabolomics starting with screening in a whole cell assay, followed by the definition of physiological response and target engagement markers and finally deciphering the mode of action.
