Abstract: Small molecule drug plasma bioanalysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS) is routinely used to quantify drug concentrations in nonclinical animal studies. The data derived from plasma quantification provides useful pharmacokinetics information to advance the understanding of small molecule drug candidate(s) to the research discovery teams. In in vivo studies, blood samples are collected, and plasma harvested at designed time points following administration to animals. The plasma bioanalysis includes a labor intensive sample preparation followed by LC-MS/MS analysis. We have introduced an automated plasma sample preparation to enhance sample preparation workflow using the Biomek i7 workstation. The sample preparation – including the preparation of standard curve, quality control samples and unknown sample are prepared within 47 min. The prepared samples are analyzed using by an ultra-high pressure liquid chromatography (UHPLC; Shimadzu LC-30 stack) system that is coupled with Sciex 6500+ triple quadruple mass spectrometer. The Biomek i7 automated workstation demonstrates a robust and reliable sample preparation for plasma derived from a range of nonclinical species. The performance of the automated sample preparation platform was verified by conducting a manual preparation followed by UHPLC-MS/MS analysis. Excellent correlation (goodness of fit, R2 = 0.997) was observed in a linear plot of the concentrations derived from the automated and manual prepared samples from at least ten different studies. The UHPLC-MS/MS has significantly reduced the sample analysis time to approx. 1 minute/sample, cut the cost by using the small size of LC column, and reduced the pollution to environment by cutting the usage of mobile phases. This combination of automated sample preparation workflow and the UHPLC-MS/MS for sample analysis helps increase the bioanalysis throughput, data turnaround time, reduce repetitive injuries from manual pipetting and reduce possible human error.
