Hepatic transport proteins play a key role in the disposition of many drugs and bile acids. Altered function of these transporters by drugs or disease may predispose patients to drug-induced liver injury (DILI). Important considerations in assessing drug interactions with hepatic transporters and bile acid-mediated DILI liability will be discussed. Preclinical tools used in drug discovery and development to identify mechanisms of interactions and prospectively predict hepatotoxic potential of new drug candidates will be highlighted.
