Description: Phase I and II metabolites especially labile glucuronide conjugations can be difficult to thoroughly characterize by CID MS/MS. In this first study we examine the use of electron fragmentation (EAD) for the characterization of Darunavir and its metabolites. From the Zeno CID MS/MS the characteristic neutral loss of m/z 176.03 is observed and no fragments that localize the glucuronide. Utilizing Zeno EAD MS/MS enables the o-glucuronide to be distinguished and localized versus the n-glucuronides. Then, microflow LC-MS/MS using a trap-and-elute set-up has been shown to deliver excellent sensitivity and selectivity for quantifying oligonucleotides. In this study, a direct injection (analytical column only) workflow was compared with the trap and elute workflow for the quantitative analysis of oligonucleotides in matrix.
