Diabetes mellitus is a metabolic disease associated with hyperglycemia, immune disorders, insulin resistance, hypertension, as well as inflammatory damage to various organs and tissues. In this study, we investigated how diabetic mellitus affects enamel and dentin formation in mice. Mandibles were dissected from 10-week-old type-2 diabetic mice (Lepr^db, #000642, Jackson laboratory) and a healthy control group. Following fixation with 10% formalin, mandibles were scanned via the Skyscan 1172 micro-CT system. Raw image reconstruction was performed using NRecon V1.4.0. and the volume and density of dentin, enamel, and pulp of the mandibular incisors were measured using CTan. Consecutive cross-sectional images of the incisors were analyzed in two anatomical sections: an apical segment and a coronal segment. The apical segment was defined as the area between the apical end of the incisor and the mesial aspect of the first molar. The coronal segment was defined as the area between the mesial aspect of the first molar and the alveolar crest of the mandibular incisor. Statistical analyses were performed using two-tailed student's t-tests. Compared with control mice, incisor enamel volume was significantly reduced in the apical segment by 55.7% in diabetic mice. Incisor enamel density was significantly decreased by 20.3% at the apical segment and by 6.5% in the coronal segment in diabetic mice. Incisor dentin volume of diabetic mice was reduced by 29.3% in the apical segment. However, dentin densities were not significantly different between control mice and diabetic mice. Diabetic mice show developmentally delayed dentin and enamel formation in incisors.
