Comprehensive Genomic Profiling of biological samples can be challenging. Inaccurate and inefficient analysis can result in inaccurate data, the cost of which can be significantly high. Formalin Fixed Paraffin Embedded (FFPE) biopsy samples are the primary sample type for assessing tumor-specific genotypes as well as expression profiles in NGS-based assays. To confidently assess sequence variants comprehensivelyDNA and RNA must be isolated from the clinical biopsy sample which includes tumor as well as possibly surrounding healthy tissue. Frequently, biopsy samples are vary considerably in size/volume and, and therefore fail to provide sufficient quantities of high qualityDNA and RNA.
In this workshop, we shall introduce Covaris® truXTRAC® FFPE Total NA Auto 96 Kit. The truXTRAC FFPE total NA Auto 96 Kit is designed for efficient and sequential extraction of total nucleic acids (RNA and DNA) from Formalin-Fixed, Paraffin-Embedded (FFPE) tissue samples using Covaris Adaptive Focused Acoustics (AFA) in a fully automated workflow.
The first step in the workflow represents excess paraffin removal using a novel Deparaffinization Solution. In the following step, after addition of Tissue Lysis Buffer, AFA-energetics® enables tissue rehydration, homogenization, and simultaneous active removal of residual paraffin from FFPE tissue samples. AFA-enhanced tissue homogenization is highly efficient for extracting macro molecules such as nucleic acids from FFPE samples. AFA in combination with a mild aqueous buffer results in increased nucleic acid yields while minimizing degradation of nucleic acids exposed at the FFPE section surface. Thus, the truXTRAC protocol results in high yields of high-quality RNA and DNA, ready for sensitive analytical methods such as next-generation sequencing (NGS) or qPCR/RT-qPCR.
