Abstract: PRISM is a cutting-edge technology for multiplexed cancer cell line viability screening to evaluate anti-cancer molecules or other perturbagens in hundreds of cancer models simultaneously. Cell lines are stably infected with unique DNA barcodes, pooled, and then co-treated for 5 days. Cells are then lysed for PCR amplification and barcode detection. Using automated liquid handlers and Luminex FlexMAP 3D technology, mRNA is captured from lysate, reverse transcribed, amplified, and detected. PRISM has adapted the Luminex detection platform’s 500 polystyrene beads by coupling to complementary oligonucleotides that are then excited by lasers to measure the PRISM barcode abundance. Using relative barcode abundance allows for the generation of cell line sensitivity signatures by comparing treatment conditions to control conditions. The PRISM sensitivity profiles are used in conjunction with genomic information to generate predictive models to identify features that correlate with sensitivity.
