Abstract: Innovations in 3D-printing have enabled rapid design-test-build cycles of advanced tissue culture devices. We have developed a high-throughput, individually-perfusable in vitro platform that replicates the 3D physiology of the blood brain barrier. Our platform uses a tri-culture of astrocytes, pericytes, and microvascular endothelial cells in a hydrogel matrix paired with microfluidics to form mature vascularized tissue in a 48-well platform. We are planning to use our platform to study blood brain barrier health and transport properties when exposed to opiates and medical countermeasures. The automated culture platform integrates fluidics into the device itself to recreate physiological shear through the gel in a user-friendly manner.
