The importance of glucuronidation is illustrated by the abundance and broad tissue distribution of the enzymes, diversity of substrates and reactions catalyzed, and number of distinctive UGTs and their genetic polymorphisms.
Their importance will be presented from regulatory and safety perspectives, along with their characteristics, examples of major glucuronides, distinctive properties of acyl-glucuronides and their significance for drug-drug interactions, factors contributing to underprediction of UGTs’ in vivo contributions, and laboratory approaches for improving these predictions.
