The utility of physiologically based pharmacokinetic modeling for food effect prediction has been discussed extensively in the scientific community. There were successful cases and cases where the model could not predict the observed food effect. Decision trees and recommendations have been proposed to improve model predictive performance. In this prestation, we will review the recent advancement and gaps in predicting the food effect and discuss what can be done with the current PBPK models.
