Dr. Stephen Hatfield is an Assistant Professor in the Department of Pharmaceutical Sciences at the New England Inflammation and Tissue Protection Institute at Northeastern University in Boston. Currently his laboratory focuses on the role of hypoxia/HIF-1α and immunosuppressive A2A Adenosine Receptor (A2AR) signaling in inflamed and cancerous tissues. Through pharmacological and gene-targeting approaches, our research aims to further our understanding of the tumor-protecting mechanisms that inhibit anti-tumor immunity. This work has led to the current understanding that tumor hypoxia (low oxygen) and the accumulation of extracellular adenosine represent a major immunosuppressive barrier that prevents immune-mediated tumor eradication (hypoxia-adenosinergic immunosuppression). Recent studies have directly implicated the blockade of A2AR and the reversal of tumor hypoxia as a feasible and translatable approach to improve the immunotherapy of cancer, providing support for ongoing clinical trials. Our research has also demonstrated the powerful immune-enhancing effects of oxygenation of the tumor microenvironment and established the industry of repurposing oxygenation agents and blood substitutes for cancer immunotherapy.
