Targeting energy metabolism is a powerful approach for pest control with established insecticides targeting several distinct mitochondrial Modes of Action. Here we investigate the effects of energy metabolism inhibitors from natural sources on Spodoptera frugiperda (fall armyworm) whole insect larvae, tissue derived cell lines and purified mitochondrial preparations with the goal to improve our molecular understanding of their molecular mechanisms. Inhibitory effects of reprentatives from natural source derived actives on all established energy metabolism targets were investigated by phenotypic assessment, quantification of energy charges as well as enzyme activity measurements. Combined with 3D analyses of the proteins structures this work provides novel tools for understanding the determinants of efficacy and selectivity of biologically active molecules.
