In this tutorial, we describe how BioLegend’s cutting-edge solutions can be used to comprehensively characterize viral- (e.g. SARS-CoV-2) and self- peptide antigen-induced immune responses. Specifically, we describe methodology for antigen-reactive T cell identification and deep characterization using the Antigen Reactive T cell kit, TotalSeq™-C Universal cocktail for Cellular Indexing of Transcriptomics and Epitopes (CITE-seq) multiomics analysis, and high-parameter spectral flow cytometry incorporating BioLegend’s Spark™ and Fire™ dyes. Furthermore, we also describe the use of LEGENDplex™ Multiplex Assays for biomarker analysis in response to antigen challenge. Together, we present a unique and comprehensive portfolio of solutions to fully characterize T cell responses to viral- and self-peptide antigens in vitro to unravel underlying mechanisms leading to pathophysiology in infectious and autoimmune diseases.
