Abstract: Objective: This study characterized a colloidal microgel for angiogenic and odontogenic differentiation of cells in the presence of cell-derived ECM proteins using a 3D culture model. Methods: Viscoelastic properties of human dental pulp were determined to compare with that of colloidal microgel (n=4). ECM proteins were extracted from dental pulp stem cells (DPSCs), and MaxGel was used as a commercially available ECM protein (n=3). DPSCs were incubated in colloidal microgels in the presence of ECM proteins, and GelMA as a bulk hydrogel (n=9/group). Cell viability was determined using 3D assay. Angiogenic properties of endothelial cells in hydrogels were determined using tubule formation assay and angiogenic marker expressions. The odontogenic differentiation of DPSCs within hydrogels under odonto/osteogenic stimulus was determined using mineralization staining, calcium and ALP assays, and odontogenic marker expressions. Results: Dental pulp showed similar viscoelastic properties with that of colloidal microgel. DPSC-derived ECM showed higher collagen and GAG than MaxGel (p< 0.05). The viability of DPSCs was similar in colloidal microgel regardless of ECM proteins, while higher cell viability, calcium deposition, and ALP activity was observed in GelMA (p< 0.05). Colloidal microgels allowed tubule-like formation by endothelial cells while no tubular formation was observed in GelMA. DPSC-derived ECM in colloidal microgel upregulated odontogenic marker expressions, while MaxGel upregulated angiogenic marker expressions (p< 0.05). Conclusions: Colloidal microgel showed angiogenic properties. GelMA showed single-cell morphology without tubule formation while improved viability and mineralization by DPSCs. The bioactivity of cell-derived ECM proteins can be modified depending on the function of the hydrogel.
