(PR66) N-Acetyl Cysteine Coated Dentin Surfaces for Enhanced Cellular Activity in Lipopolysaccharide-Stimulated Inflammatory Condition

Abstract:

Objectives: To determine the viability, migration, and mineralization of Dentin Pulp Stem Cells (DPSCs) on N-Acetyl Cysteine (NAC)-coated dentin surfaces in LPS-stimulated inflammatory conditions.

Methods: First, DPSCs viability and ALP activity were evaluated on different concentrations of NAC-coated well plates in growth medium (GM) or odontogenic medium (OM) to determine the optimum concentration of NAC. After that, cell viability and morphology were assessed in LPS-stimulated inflammatory conditions after 7 days using WST-1 assay kit and confocal laser scanning microscope (CLSM). The chemotaxis effect was tested using a transwell assay after 1 day. ALP activity was assayed after 21 days of incubation in OM. DPSCs on non-coated dentin discs and culture medium were used as controls. Data were analyzed using two-way ANOVA (n=3, assayed in triplicate).

Results: Dentin discs showed similar cellular viability compared to GM (p=0.05) with increased cellular viability in LPS-stimulated conditions (p < 0.05). The highest cell viability was observed on NAC-coated discs (p < 0.05) with similar elongated cell morphology with non-coated dentin discs. Cells showed higher migration through dentin discs than the control group regardless of the coating and LPS-condition (p < 0.05). The highest ALP activity was observed on NAC-coated dentin discs with a less pronounced effect in LPS-stimulated conditions (p < 0.05).

Conclusion: Coating dentin surfaces with N-Acetyl Cysteine enhances cellular activity in normal and LPS-stimulated environments.
