Abstract: This lecture and discussion explores basic principles of pharmacology that impact the ototoxic potential of drugs. Chemical characteristics associated with the potential for adverse effects, including ototoxicity, will be briefly reviewed. Existing drugs with known ototoxicity will be discussed and information about newer drugs with possible ototoxic effects will be introduced.

Learning Objectives:

Define and describe important pharmacokinetic parameters that potentially contribute to ototoxicity.
List common medications that have “oto-irritating” or potentially ototoxic adverse effects.
Describe the possible ototoxic adverse effects of investigational medicines.
